Yes, but the primary problem to solve is where to boot from.
If the second drive is bootable in macos, then no problem, you should be able
to just use it normally.
If the second drive is not bootable, you have to modify your bootable
original drive's partition table to include the small boot partition. Mine
is 30mb. 10mb may be enough.
The root partitition /, /home and any other partitions you want can go on the
new second drive.
